The aggregation concern is more biologically relevant than degradation for stacking - a degraded molecule is just inactive, whereas aggregate formation could potentially trigger immune responses. The current evidence on in-body stacking doesn't show the problem but it's also not extensively studied in the combined formulation context.
 
yeah the cagrisema study was the main reference point and sema turned out to be the weaker partner in that combination, making it harder to know what cagri was actually adding. the tirz plus cagri picture coming out of real world use is much more interesting. individual variation in reta stacking is real too - hard to generalize from any single study.
